Media - YEM (YEast extract - Manitol)
This is a very basic recipe used to grow rhizobium strains.
|1 liter||2 liter||4 liter||5 liter|
|Mannitol||5 g||10 g||20 g||25 g|
|Yeast extract||0.5 g||1 g||2 g||2.5 g|
|MgSO4 x 7 H20||0.2 g||0.4 g||0.8 g||1 g|
|NaCl||0.1 g||0.2 g||0.4 g||0.5 g|
|K2HPO4||0.5 g||1 g||2 g||2.5 g|
|Na Gluconate||5 g||10 g||20 g||25 g|
|add water to||1 l||2 l||4 l||5 l|
Prepare 16.6% CaCl2 solution.
Autoclave for 20 min at 120 °C
Add 1 ml of the 16.6% CaCl2 solution to 1 liter YEM medium (1:1000)
Let cool to ca. 55 °C before adding antibiotics
Store at room temperature or 4 °C
Prepare YEM as described above, but add before autoclaving 1,5 % Agar (15 g per liter)
Yeast Extract (# Y1333.1000) by Duchefa
