This is a very basic recipe used in any lab.
It is probably THE most used bacteria medium.
Costs: 0.85 euro/liter
Composition:
|
|
||||||||||||
-- | add 950 ml water | ||||||||||||
adjust the pH to 7.0 with 5 M NaOH | |||||||||||||
-- | add water to 1 liter |
Autoclave for 20 min at 120 °C
Let cool to ca. 55 °C before adding antibiotics
Store at room temperature or 4 °C
Plates:
Prepare LB as described above, but add before autoclaving:
1,5 % Agar | 15 g Agar |
Materials needed:
Tryptone (# T1332.1000) by Duchefa
Yeast Extract (# Y1333.1000) by Duchefa
Known Issues:
- I always use LB with half of the NaCl concentration and I have no clue why.
- I never ajusted the pH because I am lazy and the bacteria didn't mind it.
